【摘 要】利用高灵敏度、全自动化的方法对微量和痕量样本的DNA分型一直是法医学工作的重点与难点。焦磷酸测序技术具有实时监测、高通量、无需胶检测和荧光标记、定量性能好、结果准确和可自动化的优势。本文就焦磷酸测序化学反应测序的原理、流程及其在法医学SNP分型、建立标准化分型体系、大规模人群数据库建设及与其他常用分型技术的比较做一综述。

【Abstract】 Analyzing minute amounts of DNA is a routine challenge in forensics in part due to the poor sensitivity of the instrument and its inability to detect results from samples.Py-rosequencing method is a real- time detection, high throughout, no gel electrophoresis and good quantitative method in forensic science and population genetic areas. In this paper, we explained the theory and procedure of pyrosequencing method, detailed the meaning of constructing standard system and forensic database, and discussed the difference between pyrosequencing and other SNP genotype methods.
